Background

Prostate-specific antigen (PSA), also known as kallikrein III, seminin, semenogelase, ?-seminoprotein and P-30 antigen) is a 34 kD glycoprotein manufactured almost exclusively by the prostate gland; PSA is produced for the ejaculate where it liquifies the semen in the seminal coagulum and allows sperm to swim freely.1It is also believed to be instrumental in dissolving the cervical mucous cap, allowing the entry of sperm.2It is a serine protease enzyme, the gene of which is located on the nineteenth chromosome (19q13).3

Principle

KALLIKREIN 3 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. Human KALLIKREIN 3 specific-specific polyclonal antibodies were precoated onto 96-well plates. The human specific detection polyclonal antibodies were biotinylated. The test samples and biotinylated detection antibodies were added to the wells subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human KALLIKREIN 3 amount of sample captured in plate.
