The in vivo grade recombinant anti-mouse/human/rat/monkey/hamster/canine/bovine TGF-β mouse IgG1 kappa monoclonal antibody was produced in mammalian cells.
Clone: 1D11.16.8.
Isotype: Mouse IgG1 Kappa.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: 1D11.16.8) specifically binds to mouse/human/rat/monkey/hamster/canine/bovine TGF-β.
Applications: ELISA, flow cytometry, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the TGF-β protein.
Formulation: 0.2 μM filtered solution of 1x PBS.
Purity: >95% by SDS-PAGE under reducing conditions.

Mice received 200 μg of anti-mouse TGF-β antibody (1D11.16.8, Bio X Cell) or isotype control antibody (MOPC-21, Bio X Cell) via intraperitoneal injection 24 h prior to ischemia-reperfusion (I/R) surgery. Kidney injury was induced by clamping both renal pedicles for 26 min followed by reperfusion. Mitochondrial mass was assessed using MitoTracker Deep Red staining and flow cytometry. TGF-β blockade significantly reduced the expression of proinflammatory cytokines such as IL-6 and TNF-α. Histological analysis revealed decreased tubular necrosis in the anti-TGF-β-treated group compared to controls.

TGF-β neutralization enhances iNKT cell-mediated antitumor immunity by preserving RORγt expression and function

Mice were treated with 100 μg of anti-mouse TGF-β antibody (clone 1D11.16.8, Bio X Cell) intraperitoneally every other day starting on day 0. Tumor growth was monitored by caliper measurements twice weekly. iNKT cell activation was assessed by IFN-γ production and RORγt expression via flow cytometry. Combination therapy with iNKT cells and TGF-β blockade significantly reduced tumor burden compared to single treatments. Splenocytes were harvested on day 14 for ex vivo analysis of immune cell populations.

Anti-TGF-β antibody enhances the antitumor effect of radiotherapy in gastric cancer

Anti-TGF-β antibody (1D11.16.8, Bio X Cell) was administered intraperitoneally at 5 mg/kg twice weekly. Tumors were established by subcutaneous injection of MKN45 gastric cancer cells into BALB/c nude mice. Radiotherapy was delivered at 5 Gy per fraction for three consecutive days. The combination of TGF-β blockade and radiotherapy significantly prolonged survival compared to either treatment alone. Immunohistochemistry revealed reduced collagen deposition in the tumor microenvironment of the combination group.

TGF-β Blockade Does Not Improve Control of an Established Persistent Viral Infection

TGF-β was neutralized in vivo by administration of 250 μg anti-TGF-β (1D11.16.8, Bio X Cell) intraperitoneally every other day. Mice were infected with 2 × 10^6 PFU of LCMV clone 13 via intravenous injection. Viral titers were quantified in serum and tissues using plaque assays. CD8+ T cell responses were analyzed by tetramer staining and intracellular cytokine staining. TGF-β neutralization did not significantly enhance viral clearance in this chronic infection model.

Neutralization of TGF-β ameliorates experimental renal fibrosis

Neutralizing anti-TGF-β antibody (1D11.16.8) was administered intraperitoneally at a dose of 5 mg/kg three times per week. Renal fibrosis was induced by unilateral ureteral obstruction in C57BL/6 mice. Kidney tissue was harvested on day 7 for histological and molecular analyses. TGF-β blockade significantly reduced collagen I and fibronectin deposition in the obstructed kidney. Real-time PCR analysis showed decreased mRNA levels of TGF-β1 in the treated group.
