Flow cytometric analysis of TER-119 expression in erythroid progenitors during stress erythropoiesis

The TER-119 antibody was used in flow cytometry to identify erythroid progenitors in phenylhydrazine-induced stress erythropoiesis in mice. Bone marrow and spleen cells were harvested 48 hours post-induction, with erythroid differentiation staged by CD71 and TER-119 co-expression. Stress erythropoiesis increased TER-119+ cell frequency by 30%, and cell populations were analyzed using FlowJo software.

TER-119 staining for flow cytometry reveals erythroid maturation defects in beta-thalassemia mice

The TER-119 antibody was used in flow cytometry to assess erythroid maturation in Hbbth3/+ beta-thalassemia mice. Spleen cells were stained with CD71 and FSC to define maturation stages, and ineffective erythropoiesis was quantified by TER-119+ cell counts. Splenomegaly was measured by spleen weight, and hemoglobin levels were reduced by 25% in mutant mice.

TER-119 flow cytometry gating strategy for erythroid lineage in fetal liver

Fetal liver cells from E13.5 embryos were stained with TER-119 antibody for flow cytometric analysis of the erythroid lineage. CD71 co-staining identified early and late erythroblasts, and progenitor populations were sorted using a FACSAria. Erythroid differentiation was impaired in knockout embryos, highlighting TER-119’s utility in studying erythroid lineage development.

TER-119 and CD71 co-staining in flow cytometry for erythroid differentiation analysis

Flow cytometry using TER-119 antibody and CD71 was performed to stage erythroid differentiation in bone marrow from C57BL/6 mice. TER-119+ populations were gated into four maturation stages, and RNA was extracted for qPCR analysis of erythroid genes. Knockout mice showed a 40% reduction in mature erythrocytes, confirming TER-119’s role in differentiation analysis.

Gating strategies for TER-119+ erythroid cells in flow cytometry of hematopoietic tissues

The TER-119 antibody was used in flow cytometry to gate erythroid cells in spleen and bone marrow, with propidium iodide staining to exclude dead cells. Erythroid subsets were defined by CD71 and FSC profiles, and spleen cell suspensions were prepared by mechanical dissociation. Data were acquired on a FACSCalibur flow cytometer, validating TER-119 for erythroid cell analysis.
