Cis-regulatory evolution of the recently expanded Ly49 gene family

Ly49D (4D11; APC; Cat# 17-5782-82), Ly49E/F (CM4; PerCP; 46-5848-82), Ly49G2 (eBio4D11; FITC; Cat# 11-5781-82), Ly49H (3D10; FITC; Cat# 11-5886-82), Ly49I (YLI-90, FITC; Cat# 11-5895-85) NKG2A/C/E (20D5; FITC; Cat# 11-5896-85), NKG2AB6 (16a). We systematically profiled the epigenome of 10 Ly49 genes in 7 mouse strains, revealing that inhibitory and activating Ly49 genes are regulated by two distinct sets of cis-regulatory elements (CREs). Different Ly49 genes can be cross-regulated by shared CREs, which are polymorphic between mouse strains. These findings uncover the cis-regulatory mechanisms underlying the functional diversification of the rapidly expanded Ly49 gene family. Our study provides a framework for understanding the evolution of cis-regulation in multigene families and its impact on immune function.

Functional Characterization of Ly49+CD8 T-Cells in Both Normal Condition and During Anti-Viral Response

Ly49G (4D11), Ly49F (HBF-719), Ly49D (4E5), Ly49H (3D10), IFN? (XMG1.2), IL2 (JES6-5H4), TGF? (TW-716B4), Granzyme B (GB11), TNF? (MP6-XT22), IL10 (JES5-16E3). Ly49+CD8 T-cells were identified as CD8 lymphocytes that express Ly49 molecules detected by the available mAbs against Ly49A, Ly49C/F/I/H, Ly49G, Ly49F, Ly49C/I, or Ly49D+H according to the tested hypothesis. The next most expressed receptors are Ly49G and Ly49A, which are mostly expressed together with Ly49F (16.4% of Ly49+CD8 T-cells are Ly49F+G+, 3.2% are Ly49F+A+, and ~11.3% are Ly49F+G+A+). Ly49C and Ly49I are the least expressed inhibitory receptors among tested. Cumulatively, 26% of total Ly49+CD8 T-cell express activating Ly49D and Ly49H receptors mostly without other inhibitory Ly49 receptors.

Differential expression of the Ly49G(B6), but not the Ly49G(BALB), receptor isoform during natural killer cell reconstitution after hematopoietic stem cell transplantation

Splenic CD45.1+NK1.1+/CD122+CD3? NK cells from (A) B6 (H-2b), (B) BALB/c (H-2d), (C) BALB.B (H-2b) and (D) CB6F1 (B6xBALB/c; H-2bxd) hybrid mice were stained with FITC-anti-Ly49G (4D11). Lethally irradiated CB6F1 (H-2bxd) mice were transplanted with 5×106 of syngeneic NK and T cell depleted HSCs. Splenic CD45.1+NK1.1+CD3? NK cells from recipients were stained with FITC-anti-Ly49G (4D11) (A), FITC-Cwy-3 (Ly49GB6) and PE-AT8 (Ly49GBALB) mAbs (B) at day 14 after syngeneic HSCT and compared to adult resting mice (control). Lethally irradiated C57BL/6 (B6, H-2b, CD45.2) and BALB background mice (H-2d) mice were transplanted with 5×106 of Ly5.2 congenic (H-2b, CD45.1) and syngeneic HSCs depleted of NK and T cells, respectively. Splenic CD45.1+NK1.1+/CD122+CD3? NK cells from (A) BALB/c (H-2d), (B) BALB.B (H-2b) and (C) B6 (H-2b) mice were stained with FITC-anti-Ly49G (4D11) and PE-anti-Ly49C/I (5E6) mAbs.

DAP10 associates with Ly49 receptors but contributes minimally to their expression and function in vivo

(D) WT and DAP12-/- NK cells were cultured in IL-2 for 3 days and tested for cytolytic activity against Baf/3 cells transfected with m157 or CHO cells in the presence or absence of an anti-Ly49D or anti-Ly49H antibody. Our data demonstrate that DAP10 does associate with Ly49H and Ly49D in primary NK cells. While this association contributes slightly to cell surface expression of both receptors, it has no significant impact on Ly49H-mediated control of murine cytomegalovirus infection. (A) NK cells from WT, DAP10-/-, DAP12-/- and DAP10/DAP12-/- mice were purified from spleen and stained ex vivo or after culture in IL-2 (LAK) with anti-Ly49D and anti-Ly49H antibodies. Ly49D and Ly49H expression on gated NK1.1+ CD3- cells is shown.

Independent control of Ly49g alleles: implications for NK cell repertoire selection and tumor cell killing

Costaining with AT8 and a Ly49G(B6)-restricted Ab (Cwy-3) provides the first direct evidence that Ly49G protein is expressed from both alleles on a significant proportion of NK cells from four different types of F(1) hybrid mice. Surprisingly, the percentage of NK cells expressing both Ly49G alleles could be dramatically increased in vitro and in vivo through IL-2R- and IFN receptor-dependent signaling pathways, respectively. Unexpectedly, Ly49G(B6+) NK cells in an H-2(d), but not H-2(b), background were more likely to lyse D(d+) and Chinese hamster ovary tumor cells than Ly49G(BALB/129+) NK cells. These data indicate that the stochastic mono- and biallelic expression of divergent Ly49G alleles increases the range of MHC affinities and the functional potential in the total NK cell population of heterozygous mice. A novel murine NK cell-reactive mAb, AT8, was generated.
