Background
The rat anti-mouse GITR monoclonal antibody DTA-1 (rat IgG2b lambda) reacts with the mouse GITR protein (Glucocorticoid-Induced TNFR-Related protein or tumor necrosis factor receptor (TNFR) superfamily 18, TNFRSF 18). The DTA-1 monoclonal antibody reacts with the mouse GITR which is expressed predominantly by CD4+CD25+ T regulatory cells (Treg) and by CD25+ CD4+ CD8- thymocytes.
Our recombinant DTA-1 antibodies have a part (variable regions) or complete amino acid sequences of the rat anti-mouse GITR monoclonal antibody (hybridoma clone name or number: DTA-1).

GITR agonistic stimulation enhances the anti-tumor immune response in a mouse model of ESCC

At the end of 4-NQO or propylene glycol treatments, mice were injected intraperitoneally with 10 µg of the agonistic GITR antibody DTA-1 (BioXCell, West Hanover, NH, #BE0063) or rat IgG2b isotype control. Mice were treated every Tuesday and Friday for 2.5 weeks and were harvested the day after the last injection. For flow cytometry experiments, mice were sacrificed after 28 weeks of treatment. Based on these characteristics of GITR activity, we developed a flow cytometry panel to determine the composition of immune cells in the esophagus of 4-NQO mice treated with IgG or an anti-GITR agonist. Our flow cytometry analyses similarly demonstrated a decrease in Tregs and an increase in CTLs in mouse esophageal fractions following anti-GITR agonism, but these changes did not reach significance.

A murinized IgG2a version of rat anti-mouse GITR antibody DTA-1 was generated on the basis of published sequences. Antibodies used for flow cytometry are listed in the Supplementary Material. Expression of GITR in blood of humans, cynomolgus monkeys, and mice. The frequency of GITR+ cells on the indicated cell types from healthy human donors (n = 3–15), cancer patients (n = 7–22), cynomolgus monkey (n = 15–30), healthy mice (n = 5–10), and tumor-bearing mice (n = 5) was evaluated by flow cytometry. Naïve, TCM and TEM were FACS-sorted on the basis of the gating scheme shown, stimulated with anti-CD3 and anti-CD28 microbeads (at a bead-to-cell ratio of 1:2) in complete RPMI-1640, and expression of GITR on their surface was determined on day 2 post-activation by flow cytometry. Expression of GITR on CD4+ and CD8+ TILs obtained from non–small cell lung carcinoma (NSCLC), melanoma, and renal cell carcinoma (RCC) tumor tissues was evaluated by flow cytometry and compared with that on mouse TILs from 10 syngeneic mouse tumor models (MC38, MB49, LL/2, B16F10, TC1, RENCA, 4T1, CT26, EMT6, and CM3).

The DTA-1 monoclonal antibody specifically binds to GITR [Glucocorticoid-induced Tumor necrosis factor (TNF) receptor family-Related], a 66-70-kDa homodimer glycoprotein that is a member of the TNF receptor superfamily and is also known as TNFRSF18 and CD357. mAb DTA-1 abrogates suppression by Treg cells without affecting their proliferative response, while it is co-stimulatory for T lymphocytes that are not Treg cells. Flow cytometric analysis of CD357 (GITR) expression on mouse CD4-positive splenocytes. C57BL/6 splenocytes were stained with APC Rat anti-Mouse CD25, FITC Rat anti-Mouse CD4, and either PE-Cy™7 Rat IgG2b Isotype Control. The left panel represents the expression of CD357 (GITR) by CD25-positive/CD4-positive T lymphocytes (Treg cells), and the right panel shows CD25-negative/CD4-positive T lymphocytes. Flow cytometry was performed on a BD FACSCalibur™ flow cytometry system.
