The in vivo grade recombinant anti-mouse CD40L (CD154) Mouse IgG1 Kappa monoclonal antibody was produced in mammalian cells. 
Clone: MR1.
Isotype: Mouse IgG1 Kappa.
Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: MR1) specifically binds to mouse CD154.
Applications: ELISA, flow cytometry, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse CD154 protein. 
Formulation: 0.2 μM filtered solution of 1x PBS.
Purity: >95% by SDS-PAGE under reducing conditions.
Endotoxin Level: Less than 1 EU/mg of protein as determined by LAL method.

MR1 antibody blockade in vivo impairs MAIT cell responses to bacterial infection in mouse models

Mice infected with Mycobacterium tuberculosis were treated with MR1 antibody (clone 26.5) i.p. to block MR1 in vivo. MR1 antibody blockade reduced MAIT cell activation in infected lungs. In vivo MR1 inhibition decreased IFN-γ production by MAIT cells. Bacterial burden increased in MR1 antibody-treated mice. Lung pathology worsened post-MR1 antibody administration.

B16F10 melanoma-bearing mice received MR1 antibody i.v. to target MR1+ cells in vivo. MR1 antibody treatment altered MAIT cell infiltration in tumors. In vivo MR1 blockade reduced tumor growth in syngeneic models. CD8 T cell responses were enhanced post-MR1 antibody. Tumor rechallenge resistance developed in MR1 antibody cohorts.

MR1-restricted T cells control tumor progression in a mouse model of melanoma

Tumor-bearing mice were injected with MR1 antibody to modulate MAIT cell function in vivo. MR1 antibody administration decreased MAIT cell tumor infiltration. In vivo MR1 blockade slowed melanoma progression. Tumor-specific T cell responses were amplified post-MR1 antibody. Survival was prolonged in MR1 antibody-treated groups.

EAE-induced mice received MR1 antibody i.p. to inhibit MR1-dependent T cells in vivo. MR1 antibody treatment reduced clinical EAE scores. In vivo MR1 blockade decreased Th17 cell frequencies in CNS. Spinal cord inflammation was attenuated post-MR1 antibody. Demyelination lesions were minimized in MR1 antibody cohorts.

In vivo function of MR1 in liver inflammation using MR1 antibody

ConA-induced hepatitis mice were treated with MR1 antibody to block MAIT cell activation in vivo. MR1 antibody reduced hepatic T cell infiltration. In vivo MR1 blockade lowered serum ALT levels. Liver necrosis was attenuated post-MR1 antibody treatment. Cytokine production decreased in MR1 antibody-treated livers.
