1、A novel MyD88 inhibitor attenuates allograft rejection after heterotopic tracheal transplantation in mice.
Yang, M., et al. Transpl Immunol. 2019 Apr;53:1-6. doi: 10.1016/j.trim.2018.11.006. PMID: 30472390
The allograft recipients were treated with TJ-M2010–5, and anti-mouse CD154 (MR-1). ...However, the loss of the epithelium and airway obstruction were significantly improved in mice treated with TJ-M2010–5 combined with MR-1. ...In heterotopic tracheal transplantation models, TJ-M2010–5 combined with MR-1 could ameliorate the development of OB. ...Despite triple or quadruple immunosuppressive therapy, acute and chronic rejection continues to be the major limitation to long-term survival after lung transplantation. ...We developed the novel MyD88 molecular inhibitor TJ-M2010–5 based on the MyD88 molecular structure.

2、MR1-ligand cross-linking identifies vitamin B6 metabolites as TCR-reactive antigens.
Schmidlin, T., et al. Cell Rep Methods. 2025 Aug 18;5(8):101120. doi: 10.1016/j.crmeth.2025.101120. PMID: 40763744
Antigen presentation on the cell surface by major histocompatibility complex (MHC) class I-related protein 1 (MR1) is a vital part of mammalian immune surveillance. ...Since then, MR1 has been suggested to play a role in the immune response to bacterial infection and is now known to be a central molecule involved in immune recognition and control of infectious diseases due to its ability to present metabolites to T cells. ...K43 also performs a key role in retaining MR1 in the endoplasmic reticulum (ER) prior to ligand capture. ...These efforts have resulted in less than twenty compounds being classified as bona fide or potential MR1 ligands and T cell antigens to date, including products and derivates of the vitamin B2 and B9 pathways. ...Here, we present an MS-based method to investigate MR1-bound antigens at the cell surface of cancer cells.

3、Recognition of MR1-antigen complexes by TCR Vγ9Vδ2.
Loureiro, J. P., et al. Front Immunol. 2025 Feb 18;16:1519128. doi: 10.3389/fimmu.2025.1519128. PMID: 40040716
Rare TCR γδ cells were also shown to interact with MR1 tetramers loaded with 5-OP-RU or acetyl 6-formyl pterin (Ac-6-FP). ...These data indicated that rare T cells expressing a TCR γδ may interact with MR1 using an innate-like target recognition mechanism that is Ag-independent. ...We investigated whether TCR γδ cells may also recognize MR1-Ag complexes by an adaptive-like mechanism. ...A375 cells were tested for MR1 surface expression by flow cytometry. ...Then, MR1-coated beads were washed with PBS and used for activation assays or TCR tetramer staining experiments.

4、MR1 deficiency enhances IL-17-mediated allergic contact dermatitis.
Imahashi, N., et al. Front Immunol. 2023 Jun 20;14:1215478. doi: 10.3389/fimmu.2023.1215478. PMID: 37409131
The involvement of other subsets of innate-like T cells was also revealed in MR1-/- mice, and their relevance in ACD is discussed. ...5-OP-RU loaded MR1 tetramer was provided by the National Institute of Health Tetramer Core Facility at Emory University (Atlanta, GA, USA). ...To examine the role of MR1/MAIT cells in ACD, WT and MR1-/- mice were sensitized with DNFB in an acetone/olive oil solvent on the abdominal skin and challenged five days later on the right pinna, and the increment in thickness of the pinna in each mouse was calculated. ...In contrast, the cells of interest in the present study, MAIT cells detected by 5-OP-RU/MR1 tetramer, were reduced in DNFB-challenged pinnae in comparison with those in the vehicle control, (Figure 3C) both in terms of frequency and cell number (Figure 3D). ...The increment and reduction of each T cell subset, as compared with other immune cells among the different panels, are not evident, because the cell numbers during the pre- and post-elicitation stages of each cell number differ over several log scales.

5、Two-pore channels in MR1-dependent presentation of Mycobacterium tuberculosis infection.
Karamooz, E., et al. PLoS Pathog. 2025 Aug 4;21(8):e1013342. doi: 10.1371/journal.ppat.1013342. PMID: 40758737
MAIT cells are highly prevalent T cells that detect antigens presented on MR1. ...We have previously found that endosomal trafficking proteins are important for MR1 presentation of Mycobacterium tuberculosis (Mtb) infection. ...Our results suggest that endosomal calcium signaling and calcium-sensitive trafficking proteins are necessary for MR1 presentation of intracellular microbes. ...Mucosal Associated Invariant T cells (MAITs) are an abundant class of CD8 + T cells restricted by MR1 with a high prevalence in the peripheral blood as well as certain organs such as the lungs. ...Despite the commonality of endosomal markers between the MR1 and Mtb compartment in BEAS-2B, it is unclear whether MR1 is physically present in the Mtb compartment.
