The results showed a clear difference between the two CD137 agonist antibodies tested, clone 3H3 induced a sustained increase in T cell infiltration, proliferation and activation in the liver and spleen, whereas clone Lob12.3 (anti-mouse CD137 antibody) did not (Fig. 6B). For cell binding assays cells were incubated with primary antibodies or mAb 2 followed by detection with an anti-human Fc-488 secondary antibody (Jackson Immunoresearch). Cells isolated from tissues and blood were stained for flow cytometry with fluorochrome conjugated antibodies including those against CD4, Ki67, FoxP3, CD69, CD3, CD8 (Thermo), CD45 (BD) and fixable live/dead dye (Thermo) in the presence of Fc block (Thermo) according to manufacturer’s recommendations. TIL analysis of day 21 CT26 tumors (n=5) treated with 1 mg/kg FS120 surrogate or controls Q2D starting on day 10 for 3 doses injected intraperitoneally by flow cytometry. Tumor growth was monitored by caliper measurements twice weekly.

Optimization of 4-1BB antibody for cancer immunotherapy by balancing agonistic strength with FcγR affinity.

We observed similarly increased ALT levels in 3H3-treated naive mice (Fig. 1e, f), suggesting 3H3-induced liver toxicity is independent of tumor burden. Besides elevation of ALT in serum, we also found that immune cell infiltration in the liver of 3H3-treated mice (Fig. 1g). Our data suggested anti-4-1BB Ab-induced anti-tumor activity and liver toxicity could be separated in natural anti-4-1BB Abs. We, therefore, used LOB12.3 (anti-mouse CD137 antibody) and 3H3 Abs as tool molecules to investigate the underlying mechanism in order to gain insights for clinical development of agonistic anti-4-1BB Abs. Flow cytometry analysis showed increased CD8+ T cell infiltration in tumors treated with LOB12.3 compared to controls.

The agonistic anti-4-1BB antibody (clone Lob12.3) was administered intraperitoneally to enhance T cell responses in HPV+ tumor models. Mice were vaccinated with E6/E7 peptides on days 7 and 14 post-tumor implantation. Tumor growth was monitored by caliper measurements. Flow cytometry was used to assess CD8+ T cell infiltration using anti-CD8 (clone 53-6.7). Combination therapy led to durable tumor regression in 80% of treated mice.

Flow cytometry was performed using a BD LSRFortessa. CD8+ T cell dysfunction was assessed by LAG-3 and 4-1BB expression. Data were analyzed with FlowJo software.

Anti-4-1BB (LOB12.3) antibody was injected intraperitoneally to activate CD137 signaling in AML models. Mice were challenged with C1498 leukemia cells. Flow cytometry analysis confirmed increased CD8+ T cell activation with LOB12.3 (anti-mouse CD137 antibody) treatment. Survival was monitored for 60 days post-treatment. Combination with anti-TIM-3 enhanced long-term protection.
