Source: The anti-human/rat/fish AChR monoclonal antibody (clone: mAb35) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: mAb35) specifically binds to human/rat/fish AChR.
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human/rat/fish AChR protein.
Form of Antibody: 0.2 uM filtered solution, pH 7.4, no stabilizers or preservatives.

Treatment of passively transferred experimental autoimmune myasthenia gravis using papain

The anti-AChR mAb35 antibody was used to induce experimental autoimmune myasthenia gravis (EAMG) in 4-week-old female Lewis rats via intraperitoneal injection. Papain treatment with mAb35 reduced serum anti-AChR levels within hours, leading to mild weakness and recovery, while untreated mAb35 rats developed severe symptoms and died within 24?30 hours. Papain alone showed no side effects for up to 2 months.

The mAb35 antibody, recognizing alpha-bungarotoxin-insensitive nAChRs, was used for immunocytochemistry in guinea pig gut and pancreas. It revealed receptor clusters on Dogiel type I motorneurons and secretomotor neurons in the bowel, with internal sites in permeabilized ganglia closely associated with ChAT-immunoreactive nerve fibers.

The mAb35 antibody, targeting nAChRs, caused a time-dependent reduction in Emax for nicotine, epibatidine, and cytisine in bovine adrenal chromaffin cells, indicating a loss of nAChR-mediated secretion. Northern analysis confirmed the presence of a beta 4 subunit transcript, supporting the existence of a nAChR reserve downregulated by mAb35.

The mAb35 antibody, recognizing the alpha-subunit of muscle and neuronal nAChRs, reduced nAChR-stimulated catecholamine release in cultured chromaffin cells (IC50 ~10nM). Immunocytochemical studies confirmed specific binding to chromaffin cell sites, with no effect on non-nAChR-mediated release, establishing mAb35’s role in targeting principal adrenal nAChRs.

Preclinical evaluation of 67Cu-labeled intact and fragmented anti-colon carcinoma monoclonal antibody MAb35

The anti-carcinoembryonic antigen mAb35 and its F(ab')2 fragment, labeled with 67Cu or 131I, were evaluated in nude mice with human tumor xenografts. The 67Cu- and 131I-labeled mAb35 IgG showed similar tumor uptake and retention, while the 67Cu-labeled F(ab')2 fragment exhibited faster blood clearance than its 131I counterpart.
