Epidermal growth factor receptor (EGFR) is commonly overexpressed in NSCLC and its overexpression frequently correlates with a poor prognosis. ...There are two approaches for inhibiting EGFR signaling: to prevent ligand binding to the extracellular domain with a monoclonal antibody and to inhibit the intracellular tyrosine kinase activity with a small molecule. ...These disappointing results of EGFR tyrosine kinase inhibitors have led to increased interest in the monoclonal antibodies against EGFR because these two classes of agents may have substantially different mechanisms of action. ...To determine the absolute number of EGFR molecules per cell, we carried out a quantitative flow cytometric analysis using DAKO QIFIKIT (DakoCytomation, Copenhagen, Denmark) and EGFR monoclonal antibody (clone 528). ...The slides were preblocked with 10% normal rabbit serum at room temperature for 20 min and incubated at 4°C overnight with the primary antibody, anti-EGFR antibody (clone 31G7).
