Background:
The recombinant N901 antibody binds to the human CD56 protein, a homophilic binding glycoprotein expressed on the surface of neurons, glia and skeletal muscle. CD56 is a marker of neural lineage commitment. CD56 expression is found in the hematopoietic system, natural killer cells, and other lymphoid cells, including gamma delta (γδ) Τ cells and activated CD8+ T cells, as well as on dendritic cells. CD56 has a role in cell–cell adhesion, neurite outgrowth, synaptic plasticity, and learning and memory.

The N901 antibody (anti-human CD56, Pe-conjugated) was used in multiparameter flow cytometry to characterize immune cells in humanized patient-derived tumor mice. Staining was performed on spleen single-cell suspensions and peripheral blood using FACSCanto-II or Navios cytometers, with DuraClone protocols, to assess CD56 expression alongside other markers like CD3, CD19, and CD45 for immune profiling.

The N901 antibody (anti-human CD56) was included in antibody panels (TCR, RE ALB, CAR-T1, CAR-T2, NK1, NK2) for staining EDTA blood samples in CAR-T cell immune monitoring. Flow cytometry using a DxFLEX cytometer analyzed NK cell populations, with 100 µl whole blood stained for 15 minutes to assess CD56 expression in CAR-T therapy contexts.

The N901 antibody (anti-human CD56) was part of the EuroFlow antibody panels for standardized flow cytometric immunophenotyping of leukocytes. Used with EuroFlow SOPs on FACSCanto-II cytometers and Infinicyt software, it facilitated consistent analysis of CD56 expression in normal, reactive, and malignant leukocytes, supporting WHO-based diagnosis of hematological malignancies.

The N901 antibody (anti-human CD56, NKH-1) identified CD56 expression on leukemic cells with high NK activity in aggressive NK cell leukemia/lymphoma. Flow cytometry showed these cells lacked Leu 7 (HNK-1) and T/B-cell lineage markers, confirming a distinct third lymphoid lineage with no T beta, T gamma, or immunoglobulin gene rearrangement.
