Description

Background

Recombinant mouse anti-biotin antibodies recognize biotin (vitamin B7) attached to proteins, peptides, nucleic acids, or solid matrices, and can be offered in fluorophore and enzyme-conjugated versions.

The anti-biotin monoclonal antibody can be used as a second-step reagent in various applications for detecting biotinylated targets. The biotin-streptavidin system is widely used in immunoassay signal amplification techniques because streptavidin binds biotin with extremely high affinity.

Because biotin is naturally present in a large variety of biological samples, it may affect the results of biotin-based assays, such as reduced sensitivity, false negatives or false positives. Streptavidin/biotin blocking kit, avidin/biotin blocking system, or biotin blockers are used to block signals from endogenous avidin, biotin and biotin-binding proteins in tissues when used with biotin-based IHC detection.

Signature Fragment Ions of Biotinylated Peptides

Anti-biotin antibody-based enrichment of biotinylated peptides (e.g., BioSITe) coupled to LC?MS/MS permit large-scale detection and localization of sites of biotinylation. Peptides were separated using a 75 ?m × 15 cm PicoFrit column (New Objective) packed in-house with 1.9 ?m C18 resin (Dr. Maisch GmbH) at a flow rate of 300 nL/min. Mass spectrometry data were acquired using a Q Exactive HF mass spectrometer (Thermo Fisher Scientific) in data-dependent mode. We identified a total of 1,247 unique biotinylated peptides from BioID and APEX experiments, with an average of 3.2 biotinylation sites per peptide. The presence of biotin adducts significantly altered the fragmentation patterns, leading to the generation of characteristic signature ions that were consistently observed across different experiments.

Autocatalytic amplification of Alzheimer-associated A?42 peptide aggregation in human cerebrospinal fluid

Solutions of freshly isolated monomeric recombinant A?42 were complemented with buffer, salts, ThT, and CSF to obtain a series of samples with varying CSF concentration (0?80%) at constant A?42 concentration (3 or 5 ?M), and constant concentration of all other components: 10 ?M ThT, 20 mM HEPES/NaOH, 140 mM NaCl, 1 mM CaCl2, pH 8.0. The aggregation curves from one experiment are shown in Fig. 1a and from the other in Supplementary Fig. 2, and a power function was fitted to the resulting values where ? is the scaling exponent, c the A?42 concentration, and ? a proportionality constant. Cryo-EM images of the final aggregates formed in solutions with a total monomer concentration of 10 ?M and a 0%, b 15%, or c 66% CSF show that fibrils are clearly formed also in the presence of CSF, appearing similar to fibrils formed at the same ionic strength without CSF. In all of 15%, 32%, and 66% CSF (Fig. 6b, d, h), the aggregation is retarded relative to buffer over the entire monomer concentration range, with the delay reflected in an increase in lag time and all curves have very steep transitions. The key finding of the kinetic analysis, which is consistent across all CSF concentrations, is thus a failure to fit the data unless a secondary nucleation pathway is included in the model, implying that the secondary nucleation pathway, discovered for A?42 in pure buffer system, is a dominant microscopic mechanism for the generation of new aggregates also in CSF.

Liposomes containing pure phosphatidylcholine or a mixture of 85 mol% phosphatidylcholine and 15 mol% phosphatidylserine were incubated with or without peptide, and intrinsic tryptophan fluorescence was determined. CD (Fig. 1D) suggests that C23 is largely unstructured in solution (blue circles), even in the presence of liposomes (red circles). Fusion assays were performed on the basis of a method described previously, with the following modifications. When wild-type, full-length Drosophila Atlastin is reintroduced, normal ER morphology is restored. We produced proteoliposomes that contained increasing concentrations of the non-bilayer-forming lipid phosphatidylethanolamine (PE).
