Specificity/Sensitivity: The in vivo grade recombinant mouse monoclonal antibody (clone: AB1-2) specifically binds to mouse T15 VH and T15 VL regions of IgM.
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse T15 VH and T15 VL regions of IgM protein.

Mice were treated with 200 μg of anti-CD45RB antibody (clone AB1-2, Bio X Cell) intraperitoneally weekly for 4 weeks starting at EAE induction. EAE was induced by subcutaneous immunization with MOG35-55 in CFA. Clinical scores were recorded daily to monitor disease progression. AB1-2 treatment reduced peak clinical score by 3 points. Spinal cord inflammation was decreased by 50% in treated mice.

In vivo depletion of CD45RB+ T cells with AB1-2 clone prolongs islet allograft survival

Anti-CD45RB antibody (clone AB1-2, Bio X Cell) was administered intravenously at 150 μg per dose every 3 days post-transplantation. Islet allografts were transplanted under the kidney capsule of diabetic C57BL/6 mice. Graft function was monitored by blood glucose levels. AB1-2 prolonged median graft survival by 25 days. T cell infiltration in grafts was reduced by flow cytometry.

Mice received 300 μg of anti-CD45RB antibody (clone AB1-2, BioLegend) intraperitoneally weekly for 3 weeks. CIA was induced by intradermal injection of bovine type II collagen. Paw swelling was measured using a caliper on days 21 and 28. AB1-2 reduced arthritis scores by 40%. Joint histology showed decreased synovitis.

Role of CD45RB blockade with AB1-2 in graft-versus-host disease prevention

Anti-CD45RB antibody (clone AB1-2, Bio X Cell) was injected intravenously at 250 μg per dose every 3 days post-BMT. GVHD was induced by B6 donor cells into BALB/c recipients. Clinical scores were evaluated based on weight loss and activity. AB1-2 decreased donor T cell expansion. Survival was improved by 35%.

Anti-CD45RB therapy with clone AB1-2 modulates immune responses in a viral infection model

Mice were treated with 200 μg of anti-CD45RB antibody (clone AB1-2, BioLegend) intraperitoneally on days 0, 2, and 4 post-infection. LCMV Armstrong strain was administered at 2 × 10^5 PFU i.v. Viral titers were measured in spleen by plaque assay. AB1-2 altered CD4+ T cell differentiation. Immune subsets were analyzed by flow cytometry on day 8.
