Pharmacological characterization of PF-00547659, an anti-human MAdCAM monoclonal antibody

Dose-dependent changes in the population of LPAM+CD4+ memory T-cells in the peripheral circulation following intravenous doses of MECA-367 in the mouse. MECA-367 and PF-00547659 bound with high affinity to mouse and human MAdCAM with K(d) values of 5.1 and 16.1 pmol.L(-1) respectively and blocked the adhesion of alpha(4)beta(7) (+) leukocytes to MAdCAM with similar potency. The in vivo effects of MECA-367 and PF-00547659 on restriction of beta(7) (+) memory T lymphocytes were determined in mice and macaques, respectively, over the pharmacological dose range to confirm pharmacokinetic/pharmacodynamic relationships. PF-00547659 has potential utility in the treatment of inflammatory conditions by blocking tissue homing of activated alpha(4)beta(7) (+) leukocytes. The characterization of a rodent cross-reacting antibody as a surrogate for PF-00547659 in the search for potential pharmacological biomarkers and the in vivo effects of MECA-367 on restriction of beta(7) (+) memory T lymphocytes.

Enteral feeding preserves mucosal immunity despite in vivo MAdCAM-1 blockade of lymphocyte homing

In experiment 3, 56 mice received chow, PN, chow + MECA-367 (anti-MAdCAM-1 mAb), or chow + Isotype control Ab (IsoAb) for 5 days, followed by Peyer's patches, lamina propria, and intraepithelial lymphocyte yield with respiratory and intestinal IgA levels. MECA-367 blockade of MAdCAM-1 significantly reduced Peyer’s patch, lamina propria, and intraepithelial lymphocytes to PN levels. Diet and route affect MAdCAM-1 expression (chow > complex diet > IG-PN > PN). Decreased MAdCAM-1 expression occurred within hours of starting PN in Peyer's patches, but not mesenteric lymph nodes or the intestine, and recovered quickly with enteral refeeding. Complete MAdCAM-1 blockade reduces GALT lymphocytes to PN levels, but the chow feeding stimulus preserves IgA and early antibacterial resistance, implying the existence of non-MAdCAM-1 mechanisms to preserve mucosal immunity.

An integrin alpha4beta7•IgG heterodimeric chimera binds to MAdCAM-1 on high endothelial venules in gut-associated lymphoid tissue

Mouse tissue sections of PPs, MLNs, and PLNs were probed with MECA-367 antibody, recognizing mouse MAdCAM-1 (upper panels) or α4β7•IgG in the presence (lower panels) or absence (middle panels) of EDTA. α4β7•IgG binding to mouse HEVs in situ. The product purified by Protein G-Sepharose was identified as authentic MAdCAM-1-Fc by the anti-MAdCAM-1 monoclonal antibody (MoAb) MECA-367 using Western blot and ELISA analysis. α4β7•IgG binding to CHO cells expressing MAdCAM-1 and VCAM-1, but not ICAM-1, is significantly higher than to mock-transfected control cells, and α4β7•IgG binding to CHO cells expressing MAdCAM-1 is higher than to any other CHO transfectants, with high statistical significance (p < 0.01 vs VCAM-1, p < 0.001 vs ICAM-1). Binding was enhanced by Mn(++)-induced integrin activation, and specifically blocked by anti-integrin alpha 4 subunit and anti-MAdCAM-1 MoAbs.

Inflammatory cytokines induce MAdCAM-1 in murine hepatic endothelial cells and mediate alpha-4 beta-7 integrin dependent lymphocyte endothelial adhesion in vitro

The increased adhesion induced by TNF-α was significantly reduced to 84.8 ± 3.99% of maximal adhesion by pre-/co-treatment of HEC with the blocking mouse MAdCAM-1 Ab 'MECA-367'(10 ug/ml). TNF-α stimulates MAdCAM-1 expression in a dose-and time- dependent manner. MAdCAM-1 is a major determinant of leukocyte recruitment in chronic inflammation and is expressed by HEC in response to IL-1 beta and TNF-alpha. This system may provide a useful model for studying inflammatory mechanisms in liver disease and help determine if controlled MAdCAM-1 expression might influence inflammation in liver disease. Confluent HEC endothelial cells were incubated for 24 h with increasing concentrations of mouse TNF-α (as indicated), and MAdCAM-1 induction (measured as western blotting density), expressed as a percent of the maximum, (set as the scan density in 24 h, 20 ng/ml TNF-α treated HEC, [100%]).

Construction and adhesive properties of a soluble MadCAM-1-Fc chimera expressed in a baculovirus system: phylogenetic conservation of receptor-ligand interaction

The product purified by Protein G-Sepharose was identified as authentic MAdCAM-1-Fc by the anti-MAdCAM-1 monoclonal antibody (MoAb) MECA-367 using Western blot and ELISA analysis. When immobilized on glass it was fully functional in supporting the binding of mouse alpha 4 beta 1+ alpha 4 beta 7+ mesenteric lymph node lymphocytes, and the alpha 4 beta 1- alpha 4 beta 7+ TK1 T cell lymphoma. Binding was enhanced by Mn(++)-induced integrin activation, and specifically blocked by anti-integrin alpha 4 subunit and anti-MAdCAM-1 MoAbs. Binding was blocked by pretreatment of cells with sodium azide, and EDTA, indicating that binding is an energy-dependent process which requires divalent cations. Thus the mouse MAdCAM-1-Fc chimera produced in insect cells retains certain functional properties that typify the native receptor, and should be valuable in analysing the role of MAdCAM-1 in lymphocyte recirculation and emigration.
