Briefly, frozen sections (6 μm) were fixed with a 1:1 mix of acetone/ethanol(100%) and stained with Abs against B220 (RA3-632, BD Bioscience), CD3 (145-2C11, Ebiosciences), MAdCAM-1 (MECA-367, BioXcell), CD209b/SIGN-R1 (eBio22D1, Ebiosciences), NKp46 (polyclonal, R&D systems), and CD169 (MOMA-1, AbDSerotec). ...As the extent of viral control differs in MA/My and Tg1 spleens (Fig 4), we considered that apparent differences in splenic SLO integrity could be due to the variance in viral titers. ...Abs and isotype controls for flow cytometry were purchased from BD Biosciences, Biolegend, and R&D Systems. ...Spleen and liver gDNA samples were prepared using a Gentraprep kit. ...We gratefully acknowledge the contribution of DartMouse to genome-wide genotyping performed in this study.

Effector lymphocyte-induced lymph node-like vasculature enables naive T-cell entry into tumours and enhanced anti-tumour immunity.

In both tumour sites, PNAd+ vessels coexpressed MAdCAM-1 and VCAM-1 ...Naive OT-I cells were transferred into WT mice with i.p. B16-OVA tumours pre-treated with anti-MAdCAM-1 antibody or Rat IgG isotype control (h), or anti-PNAd antibody or Rat IgM isotype control (i). ...To determine whether infiltration was mediated by L-selectin binding to MAdCAM-1 or PNAd, we transferred OT-I cells into i.p. tumour-bearing mice pretreated to block luminally expressed molecules with injection of either anti-MAdCAM-1, anti-PNAd or isotype control. ...MAdCAM-1 blockade reduced infiltration into Peyer’s patches but had no effect on tumour infiltration. ...MAdCAM-1 did not contribute to naive T-cell infiltration, however, perhaps because it is not properly posttranslationally modified to serve as an L-selectin ligand.
