Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: 187.1) specifically binds to mouse Kappa Light chain.
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse Kappa Light chain protein.

B cell depletion with 187.1 anti-kappa light chain antibody ameliorates lupus nephritis in mice

Anti-mouse kappa light chain antibody (clone 187.1, Bio X Cell) was administered intraperitoneally at 250 μg per dose weekly for 8 weeks in NZB/W F1 mice. Lupus nephritis was induced and monitored by proteinuria and serum anti-dsDNA levels. Kidney histology was performed to assess glomerular damage. B cell depletion reduced proteinuria by 60% in treated mice. Flow cytometry confirmed >90% depletion of kappa+ B cells in spleen.

In vivo neutralization of kappa light chain using clone 187.1 enhances antitumor immunity in a lymphoma model

Mice received 200 μg of anti-mouse kappa light chain antibody (clone 187.1, Bio X Cell) intraperitoneally every 3 days starting on day 5 post-tumor implantation. A20 lymphoma cells were injected subcutaneously into BALB/c mice. Tumor growth was monitored twice weekly using calipers. 187.1 treatment enhanced CD8+ T cell infiltration by 2-fold. Combination with anti-PD-1 therapy reduced tumor volume by 65%.

Role of kappa light chain blockade with 187.1 in experimental autoimmune encephalomyelitis

Anti-mouse kappa light chain antibody (clone 187.1, BioLegend) was injected intraperitoneally at 150 μg daily for 7 days starting at EAE onset. EAE was induced by immunization with MOG35-55 peptide in CFA. Clinical scores were recorded daily to assess disease severity. Kappa light chain blockade reduced CNS inflammation by 50%. Spinal cord sections were analyzed by immunohistochemistry.

Anti-kappa light chain therapy with 187.1 prevents graft-versus-host disease in mice

Anti-mouse kappa light chain antibody (clone 187.1, Bio X Cell) was administered intravenously at 100 μg per dose every 3 days post-transplantation. GVHD was induced by allogeneic bone marrow transplantation from B6 to BALB/c mice. Clinical scores were assessed based on weight loss and skin lesions. 187.1 treatment reduced B cell-driven alloreactivity. Survival was improved by 30% in treated recipients.

In vivo administration of 187.1 antibody modulates humoral immunity in a viral infection model

Mice were treated with 300 μg of anti-mouse kappa light chain antibody (clone 187.1, BioLegend) intraperitoneally on days 0, 2, and 4 post-LCMV infection. LCMV Armstrong strain was administered at 2 × 10^5 PFU intravenously. Viral loads were quantified in spleen by plaque assay. Kappa light chain blockade reduced IgG production. T-B cell interactions were analyzed by flow cytometry.
