BLOC-2 targets recycling endosomal tubules to melanosomes for cargo delivery.
Dennis, M. K., et al. J Cell Biol. 2015 May 25;209(4):563-77. doi: 10.1083/jcb.201410026. PMID: 26008744
In BLOC-2?deficient melanocytes, the melanosomal protein TYRP1 was largely depleted from pigment granules and underwent accelerated recycling from endosomes to the plasma membrane and to the Golgi. ...Other melanosome cargoes, such as TYR-related protein-1 (TYRP1) and oculocutaneous albinism type 2 (OCA2), and a smaller cohort of TYR, exploit a distinct pathway that requires BLOC-1 for cargo exit from vacuolar early endosomes. ...Besides the aberrant localization to early endosomes and the Golgi, TYRP1 levels at the plasma membrane?as assessed by flow cytometry?were increased (by 3.5?5-fold) in BLOC-2 ? and BLOC-2 ?C relative to BLOC-2 R cells. ...To directly assess TYRP1 recycling to the Golgi, we used FRAP after expression of TYRP1-EGFP. ...Labeling was particularly evident on tubules that emanated from vacuolar endosomes and that closely apposed and/or contacted melanosomes, at least some of which were associated with AP-1 and/or contained TYRP1.

For eumelanins which give rise to brown or black pigment (comprising DHICA-melanin and DHI-melanin), TRP-1 (tyrosinase related protein-1) and TRP-2 (tyrosine related protein-2, or DCT for DOPA-chrome tautomerase) are involved. ...To detect proteins involved in melanogenesis, primary monoclonal antibodies (MoAb), Ta99 specific to human TRP-1 (Signet, Dedham, MA, USA) and T311 specific to human Tyrosinase (Novocastra, Newcastle, UK) were used. ...For fluorescent immunochemistry, Ta99 (dilution 1/50) which recognizes TRP-1 was applied to cryosections for 30 min at room temperature (RT). ...Primary monoclonal antibody (MoAb) TA99 specific to TRP-1 (Signet) was applied (dilution 1/100) for 60 min at RT. ...TRP-1 staining (red) of cross-sections shows melanocytes positioned at the basal layer of the epidermis in both 3D systems.
