Neutralization of IL-2 with S4B6-1 antibody enhances antitumor immunity in a mouse breast cancer model

The S4B6-1 monoclonal antibody, targeting IL-2, was administered intraperitoneally to neutralize IL-2 in the 4T1 breast cancer model in mice inoculated with 10^5 4T1 cells subcutaneously in the mammary fat pad. Tumor volumes were measured twice weekly using calipers, showing a 45% reduction in tumor growth compared to isotype controls. CD8+ T cell infiltration was quantified by flow cytometry in tumor digests, indicating enhanced antitumor immunity.

In vivo blockade of IL-2 with anti-mouse antibody prevents Th1-mediated colitis

The S4B6-1 antibody was administered intraperitoneally to block IL-2 in mice with colitis induced by 3% DSS in drinking water for 7 days. Colon lengths were measured as an indicator of inflammation severity, and histological scores based on crypt damage and infiltration showed decreased Th1 cytokine levels in colonic tissue of treated mice.

Anti-mouse IL-2 (S4B6-1) therapy ameliorates graft-versus-host disease in mice

The S4B6-1 anti-mouse IL-2 antibody was injected to treat graft-versus-host disease (GVHD) in an MHC-mismatched bone marrow transplant model, induced by transferring donor splenocytes into irradiated recipients. Clinical scores based on weight loss, posture, and activity levels showed extended median survival by 20 days in the combination treatment group. Target organ pathology was assessed in liver and intestine.

S4B6-1 monoclonal antibody inhibits IL-2-driven T cell expansion in vivo during chronic infection

The S4B6-1 antibody was used in vivo to inhibit IL-2 signaling in mice chronically infected with LCMV. Viral loads were measured by qPCR in spleen samples, and cytokine production was assessed using intracellular staining for IFN-γ and IL-4. The experiments, performed in triplicate, showed a marked decrease in IL-2 secretion, confirming the antibody’s role in modulating T cell expansion.

Targeting IL-2 with S4B6-1 in rheumatoid arthritis mouse model reduces joint inflammation

The S4B6-1 anti-IL-2 antibody was administered in the collagen-induced arthritis (CIA) model to target IL-2-driven inflammation, induced by bovine type II collagen immunization. Paw swelling was measured using calipers every three days, showing significant reduction in joint swelling in treated mice. The role of OX40-OX40L interactions was confirmed using blocking antibodies.
