Source: The anti-mouse/human Mac-2 (Galectin-3) monoclonal antibody (clone: M3/38.1.2.8HL.2) was produced in mammalian cells.
Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: M3/38.1.2.8HL.2) specifically binds to mouse/human Mac-2 (Galectin-3).
Applications: ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the mouse/human Mac-2 (Galectin-3) protein.

Effective rational humanization of a PASylated anti-galectin-3 Fab for the sensitive PET imaging of thyroid cancer in vivo

The rat monoclonal antibody M3/38 (M3/38.1.2.8HL.2 clone) was used to generate a chimeric Fab for in vivo targeting of human galectin-3 in thyroid cancer xenografts. PASylation of the anti-galectin-3 Fab extended serum half-life and improved tumor uptake in vivo. In vivo PET imaging with 89Zr-labeled PASylated Fab showed specific accumulation in galectin-3-positive tumors. Biodistribution studies in mice confirmed low off-target binding post-Fab administration. Tumor-to-muscle ratios were significantly higher in galectin-3-expressing models.

CD8+ TIL treated with an anti-galectin-3 antibody (M3/38 clone) had increased IFN-? secretion in vivo. Tumor-bearing mice vaccinated with tumor antigen received GCS-100 injections to disrupt galectin-3 in vivo. In vivo GCS-100 treatment led to tumor rejection in 50% of vaccinated mice. Galectin-3 blockade enhanced T cell mobility and effector function in the tumor microenvironment. Non-vaccinated mice showed no response to GCS-100 alone in vivo.

Chronic upregulation of activated microglia immunoreactive for galectin-3/Mac-2 and nerve growth factor following diffuse axonal injury

Activated microglia immunoreactive for galectin-3/Mac-2 were quantified in vivo after diffuse axonal injury. Galectin-3/Mac-2+ microglia were most abundant 1 day post-injury in mouse brain tissue. In vivo levels of galectin-3/Mac-2+ cells remained elevated at 30 days after injury. Colocalization with NGF was observed in galectin-3/Mac-2+ microglia in vivo. Sham-injured mice showed minimal galectin-3/Mac-2 immunoreactivity.

Compartmentalized reactivity of M3/38 (anti-Mac-2) and M3/84 (anti-Mac-3) in the uterus of pregnant mice

Cryostat sections of uteri from pregnant mice were probed with M3/38 (anti-Mac-2) antibody in vivo. M3/38 reacted with decidual cells surrounding the embryo on day 8 of pregnancy. By day 15, placenta and decidua basalis were intensely positive for M3/38 in vivo. Embryo tissue remained negative for M3/38 immunoreactivity on day 15. Uterine leucocytes showed strong M3/38 staining throughout pregnancy.

Thioglycollate-elicited macrophages were labeled in vivo with 125I-M3/38 antibody. M3/38 antibody recognized Mac-2 antigen on macrophage surfaces in vivo. Saturation binding showed 1.7 X 10^5 Mac-2 sites per macrophage in vivo. Co-precipitation confirmed M3/38 and M3/31 bind the same Mac-2 molecule in vivo. Mac-2 was synthesized by macrophages as detected by [35S]-methionine labeling in vivo.
