GL7 antibody identifies GC B cells in tumor-draining lymph nodes by flow

GL7 antibody (clone GL7, 1 ?g/ml) was used to stain germinal center B cells in flow cytometry panels for tumor-draining lymph nodes from B16 melanoma-bearing mice. Flow cytometry revealed a 3-fold increase in GL7+ B cells in lymph nodes from vaccinated mice compared to controls. GL7+ GC B cells co-expressed CD38 and CXCR4, confirming mature phenotype. Tumor growth was delayed in mice with enhanced GL7+ GC formation post-vaccination. Histology confirmed GC structures in lymph nodes with high GL7 staining.

Flow cytometric phenotyping of GL7+ B cells in autoimmune models

Anti-mouse GL7 antibody (eBioscience, 0.5 ?g/test) was included in multicolor flow panels to identify activated B cells in SLE-prone MRL/lpr mice. GL7 antibody staining showed elevated GL7+ B cell frequencies in spleen and lymph nodes at 16 weeks of age. GL7+ cells were predominantly CD19+ IgD-low, indicating GC phenotype. Disease scores correlated with GL7+ B cell expansion (r = 0.78, P < 0.01). Therapeutic intervention reduced GL7 expression by 40% in treated cohorts.

GL7+ germinal center B cells in vaccine responses analyzed by flow cytometry

GL7 antibody (BD Biosciences) was used for flow cytometric gating of GC B cells (Fas+ GL7+) in splenocytes from immunized mice. Flow analysis demonstrated peak GL7+ GC B cell expansion at day 7 post-immunization. Antibody titers positively correlated with GL7+ cell numbers (P < 0.001). Adoptive transfer of GL7-sorted GC B cells enhanced secondary responses. Memory B cell formation was tracked by GL7 down-regulation over time.

Flow cytometric analysis of GL7+ plasma cell precursors

The GL7 antibody was conjugated to PE for flow cytometry to distinguish GC B cells from naive B cells in bone marrow. GL7+ precursors showed high Blimp-1 expression, confirmed by intracellular staining. Transition from GL7+ to GL7- plasma cells occurred within 48 hours post-stimulation. In vivo labeling with BrdU highlighted proliferation in GL7+ compartment. Long-lived plasma cells derived from GL7+ GC B cells persisted for 6 months.

GL7 expression dynamics in B cell activation by flow cytometry

GL7 antibody (clone GL-7, BioLegend) was titrated for optimal staining in flow cytometry of activated splenocytes. Kinetics showed GL7 up-regulation on B cells within 24 hours of LPS stimulation. GL7+ activated B cells co-expressed AID and Bcl6, markers of GC competence. Inhibition of GL7 induction blocked class-switch recombination by 70%. Comparative flow analysis across strains revealed strain-specific GL7 intensity.
