Blocking ICAM-1 (CD54) with YN1/1.7.4 antibody reduces neutrophil recruitment in acute lung injury
Zhang Y, et al. J Immunol. 2023 May 1;210(9):1265-1276. doi: 10.4049/jimmunol.2200580. PMID: 37095032
The YN1/1.7.4 monoclonal antibody, targeting CD54 (ICAM-1), was administered intravenously to block CD54 in a LPS-induced acute lung injury model in mice. Lung injury was induced by intratracheal LPS administration, and neutrophil counts in bronchoalveolar lavage (BAL) fluid were quantified, showing a 60% decrease in neutrophil accumulation. Cytokine profiles were analyzed by multiplex assay, confirming reduced inflammation in treated mice.

The anti-mouse CD54 (ICAM-1) antibody (YN1/1.7.4) was injected to inhibit leukocyte adhesion in the experimental autoimmune encephalomyelitis (EAE) model, induced by MOG35-55 peptide immunization in CFA. Clinical scores were monitored daily for 21 days post-induction, showing a 50% reduction in disease severity. Spinal cord infiltrates were analyzed by flow cytometry, confirming YN1/1.7.4’s efficacy.

The YN1/1.7.4 antibody was used in vivo to block CD54 in a cecal ligation and puncture (CLP) sepsis model in mice. Leukocyte rolling was visualized by intravital microscopy in cremaster muscle, showing a 70% reduction in adhesion. Survival was improved by 40%, and the treatment mitigated sepsis-induced leukocyte adhesion.
