Background:
The recombinant NCI m971 antibody binds to the human Siglec-2/CD22 protein, a sugar binding transmembrane protein, which specifically binds sialic acid with an immunoglobulin (Ig) domain located at its N-terminus. CD22 functions as an inhibitory receptor for B cell receptor (BCR) signaling. It is also involved in the B cell trafficking to Peyer's patches in mice. In mice, it has been shown that CD22 blockade restores homeostatic microglial phagocytosis in aging brains.

Intracellular staining of 300,000 fixed cells involved permeabilization and blocking to minimize non-specific binding, with stain index and MFI ratios confirming optimal staining efficiency for α-synuclein detection.

The NCI m971 antibody (NEO-201), conjugated to Pacific Blue or PE, was used in flow cytometry to analyze tumor antigen expression on ASPC-1, BxPC3, CFPAC-1, and LS174T cell lines, showing overlap with CEACAM5 and CEACAM6. ELISA confirmed binding to recombinant CEACAM5 and CEACAM6 but not CEACAM1 or CEACAM8, with IHC verification except in PEO1 cells.

The NCI m971 antibody’s Fab fragment bound the CD22 d7 domain with low nanomolar affinity (25 nM by BLI, comparable to 75 nM by flow cytometry). Cocrystal structures showed a preconfigured paratope (RMSD 0.33 Å), with HCDRs 1–3 and LCDRs 1–2 interacting with CD22’s C β-strand and loops, unaffected by N-linked glycosylation.

The NCI m971 antibody (NCRC-23, anti-carcinoembryonic antigen) was evaluated by flow cytometry, recognizing 65% of large colorectal tumor cells with a mean antigen density of 4.9 × 10^5 sites/cell. Combinations with other antibodies improved targeting coverage, reducing tumor recurrence risk in primary tumor samples.
