Background

Neural/glial antigen 2 (NG2), also known as chondroitin sulfate proteoglycan 4 (CSPG4), melanoma chondroitin sulfate proteoglycan (MCSP), or high molecular weight melanoma-associated antigen (HMW-MAA), is a cell-surface transmembrane proteoglycan. Its approximately 250 kDa N-linked core glycoprotein can carry chondroitin sulfate chains, yielding a proteoglycan form exceeding 450 kDa. NG2/CSPG4 is expressed on oligodendrocyte precursor cells, glioma cells, and many malignant melanomas, as well as selected other tumors. It contributes to cell–substratum interactions and is involved in tumor cell proliferation, adhesion, migration, and invasion. NG2/CSPG4 also functions as a coreceptor for α4β1 integrin, making it a relevant target for studies of tumor biology and neural development.

The 9.2.27 monoclonal antibody specifically recognizes human NG2/CSPG4. The original clone was generated by immunizing BALB/c mice with an extract of human M14 melanoma cells and recognizes the core glycoprotein of the melanoma-associated chondroitin sulfate proteoglycan. Clone 9.2.27 has been used in flow cytometry, immunofluorescence/immunocytochemistry, immunoprecipitation, and immunohistochemistry to detect and characterize NG2/CSPG4-expressing cells. It has also been investigated in functional studies of melanoma cell interactions and tumor growth, providing a research tool for examining NG2/CSPG4 expression and its biological roles.
