Specificity/Sensitivity: The in vivo grade recombinant rat monoclonal antibody (clone: JES3-19F1.1.1) specifically binds to human IL-10.
Applications:ELISA, neutralization, functional assays such as bioanalytical PK and ADA assays, and those assays for studying biological pathways affected by the human IL-10 protein.

The anti-IL-10R antibody (clone JES3-19F1.1.1) was used to block IL-10 signaling in the tumor microenvironment of a multiple myeloma mouse model, significantly inhibiting tumor growth in combination therapy. Flow cytometry showed increased M1-like macrophage polarization, and survival curves indicated prolonged median survival compared to controls.

The JES3-19F1.1.1 anti-IL-10R antibody, administered intraperitoneally, elevated ceramide levels in colonic tissue of colitis-induced mice. Inflammatory scores decreased with sphingolipid modulation, and gene expression profiling revealed upregulated pathways in treated animals, indicating a role in limiting inflammation.

IL-10R blockade during chronic schistosomiasis mansoni results in the loss of B cells from the liver and the development of severe pulmonary disease

The JES3-19F1.1.1 antibody neutralized IL-10R during chronic Schistosoma mansoni infection in mice, leading to hepatic B cell depletion 4 weeks post-treatment. Pulmonary inflammation scores increased significantly, with histopathology showing granuloma formation in the lungs of treated mice.

IL-10 restricts activation-induced death of NK cells during acute murine cytomegalovirus infection

The JES3-19F1.1.1 anti-IL-10R antibody, injected in mice infected with murine cytomegalovirus, reduced viral titers in the spleen and decreased NK cell apoptosis rates. Cytokine profiles shifted towards pro-inflammatory responses, highlighting the antibody’s role in modulating NK cell survival.

IL-10 and TGF-beta redundantly protect against severe liver injury and mortality during acute schistosomiasis

Combined JES3-19F1.1.1 and anti-TGF-β antibodies in acute schistosomiasis mouse models reduced liver fibrosis and mortality from 90% to 40%. Liver histopathology showed smaller granulomas in treated cohorts, demonstrating the protective role of dual IL-10R and TGF-β blockade.
