The anti-human FAP clone FAP5 (BD Biosciences) stained cancer-associated fibroblasts (CAFs) from colorectal cancer samples, showing 85% positivity. Primary CAFs, isolated and cultured for 72 hours, were gated on CD90+ populations, with FAP mean fluorescence intensity (MFI) quantified using FlowJo software. Isotype controls confirmed FAP5 binding specificity.

The FAP5 antibody, conjugated to PE, was used in an 8-color flow cytometry panel to detect FAP on lung stromal cells from non-small cell lung cancer samples. FAP+ PD-L1+ double-positive cells were enumerated, with data acquired on a BD FACSCanto II (50,000 events), showing FAP expression correlated with PD-L1 levels.

Detection of soluble FAP in serum by flow cytometry-based assay using FAP5 capture antibody

The FAP5 antibody served as a capture reagent in a flow cytometry bead-based assay, detecting soluble FAP in serum from cancer patients (diluted 1:10) with a sensitivity of 0.05 ng/mL. Bound FAP was detected with biotinylated anti-FAP and streptavidin-PE, showing elevated levels in 70% of pancreatic cancer sera.
