BP005447: Human PD-1/PDCD1 Protein with the C-His Tag
Source: The recombinant human PD-1/PDCD1 protein, the PD1 ECD (amino acids Leu25-Gln167) fused with a His-tag at the C-terminus, was produced in HEK 293 cells.
Molecular Weight: The recombinant human PD-1/PDCD1 protein with the C-His tag has a predicted MW of 17 kDa. Due to glycosylation, the protein migrates to 38-50 kDa based on Tris-Bis PAGE result.
Activity: The recombinant human PD-L1 with the C-hFc Tag captured on CM5 Chip via Protein A can bind to the recombinant human PD-1 with the C-His Tag with an affinity constant of 2.2 uM as determined in SPR assay (Biacore T200).
Purity: >95% by SEC-HPLC and SDS-PAGE under reducing conditions.
Endotoxin Level: Less than 1 EU per ug of protein as determined by LAL method.
Formulation: Lyophilized from 0.22 um filtered solution in PBS (pH 7.4). Normally 8% trehalose is added as protectant before lyophilization.
Reconstitution: It is recommended to reconstitute the lyophilized recombinant human PD-1/PDCD1 protein in sterile H2O to a protein concentration not less than 100 ug/ml. Centrifuge the tube before opening.

Activity: The EC50 of the recombinant human PD-L1 protein with the mFc tag to the immobilized human PD-1/PDCD1 with the C-hFc tag at 1 ug/ml (100 ul/Well) was 0.1 ug/ml determined by ELISA. The recombinant human PD-1 with the C-hFc Tag captured on CM5 Chip via Protein A can bind to the biotinylated recombinant human PD-L2 with the C-His-Avi Tag with an affinity constant of 0.344 uM as determined in SPR assay (Biacore T200).
