Description

Post-translational modifications such as glycosylation, deamidation, isomerization, and oxidation have the potential to reduce affinity/potency, stability, and homogeneity of an antibody, resulting in complications to downstream process development. Algorithms has been developed to identify sequences within antibody variable regions that could present potential liabilities to product quality. This can allow the investigator to select antibodies for further development that lack these potential liabilities, or to assess whether they can be eliminated early in the antibody engineering process.

General procedure of the antibody sequence liability identification service:

The antibody variable regions of both heavy chain (VH) and light chain (VL) are analyzed for liabilities including, but not limited to N-linked glycosylation, deamidation, isomerization, pyroglutamate formation, and oxidation.

Information required from clients:

Amino acid sequence of mature heavy and light chain variable regions (VH and VL).

Deliverable:

A. Report identifying potential sequence liabilities.
