The 4G One-step RT-PCR Kit is optimized to do reverse transcription (RT) and PCR in a single tube. Except RNA samples and primers, the 4G One-step RT-PCR kit contains all components of RT and PCR mixed in one tube before starting the reaction and thus, RT and PCR are carried out sequentially in the same tube. In the 4G One-step RT-PCR kit, the reverse transcriptase, hot-start Taq DNA polymerase, and 2x RT-PCR buffer are all optimized for both the RT and PCR steps. The 4G One-step RT-PCR kit is specially designed for end-point PCR detection using RNA as a template (such as RNA virus). Using gene-specific primers (GSP), reverse transcription and PCR reactions are completed in one tube, no additional tube opening/pipetting operations are required, which improves detection throughput and reduces the risk of contamination. Integrating the superior performance of 4G Reverse Transcriptase and hot-start Taq DNA Polymerase, with an optimized buffer system, the detection sensitivity of the 4G One Step RT-PCR Kit can reach 1 pg total RNA and can be amplified Fragments up to 10 kb. The kit is provided in a convenient Master Mix format. The 2 x One-step Mix contains optimized buffer system, dNTP and loading dye; One-step Enzyme Mix contains optimized ratio of 4G Reverse Transcriptase, Murine RNase inhibitor, and hot start Taq DNA polymerase. If the blue dye is added to the 4G One-step RT-PCR Kit, PCR products can be directly loaded for electrophoresis, which greatly saves time and labor.
The PCR reagent concentrations, reaction conditions, and PCR program parameters should be determined experimentally by the investigator.
