MB102: High Efficiency DH5a Chemically Competent Cells
High Efficiency chemically competent E. coli DH5a cells offer an efficient and economical approach for DNA transformation, and can be used in a wide array of molecular biology applications. Its high competency ensures success in challenging cloning projects, and unlike electrocompetent cells, High Efficiency chemically competent E. coli DH5a cells can tolerate high salt concentration in DNA solution, greatly simplifying transformation process.
Genotype: fhuA2 (argF-lacZ)U169 phoA glnV44 ?80 ?(lacZ)M15 gyrA96 recA1 relA1 endA1 thi-1 hsdR17.
Shipping: on dry ice.
Storage: competent cells should be stored at -80°C. Thaw on ice only before use. Do not re-freeze.
Standard protocol (recommended when you need maximum number of colonies):
1. Thaw cells on ice;
2. Incubate DNA with cells in 0.5 or 1.5 ml microtubes on ice for 30 minutes;
3. Heat shock tubes at 42°C for 30 seconds;
4. Recover the cells in 0.5 ml SOC medium or LB medium at 37°C for 1 hour;
5. Plate cells on selection plates.
Quick protocol for ampicillin selection only (recommended when time is more important than number of colonies):
1. Thaw cells on ice;
2. Incubate DNA with cells in 0.5 or 1.5 ml microtubes at room temperature;
3. Plate cells on selection plates.
