Description

Capillary Isoelectric Focusing (cIEF), measures the isoelectric point and charge variation of a sample. Separating proteins or antibodies based on their net charges can help evaluate a product/lot based on charge differences. Capillary electrophoresis (CE)-based separation techniques offer several advantages over conventional electrophoretic separation methods, including superior resolution/efficiency/speed, reproducibility and enhanced quantitation capabilities. By combining CE-based separation techniques with IEF, the PI of a protein can be measured with a high degree of accuracy. cIEF can identify the presence of charge heterogeneity, which can be caused by post-translational modifications such as deamidation, sialylation and N-terminal pyroglutamic acid formation.

We use the iCE3 with Alcott 720NV Autosampler to measure the pI of your sample. This Protein Simple instrument offers accurate injection combined with simple start up and maintenance. On-board sample preparation eliminates protein stability concerns by mixing ampholytes, pI markers and methylcellulose just prior to injection. Multiple tray options include 48 standard sample vials and 96-well plate compatibility.

Service of accurate pI analysis by cIEF includes:

A. Samples are analyzed in their native (undenatured) form on the cIEF instrument.
B. The retention time of the test sample is compared to the standard to calculate the pI of each isoform and to identify the presence of charge heterogeneity, if applicable.

Service of accurate pI analysis by cIEF after deglycosylation includes:

A. Samples are deglycosylated and analyzed in their native (undenatured) form on the cIEF instrument.
B. The retention time of the test sample is compared to the standard to calculate the pI of each isoform and to identify the presence of charge heterogeneity, if applicable.

Deliverable information:
- Report showing accurate isoelectric points of each isoform.
- Charge heterogeneity profiling.
- Comparability assessment (if more than one protein submitted).

Sample treatment: native or partially denatured condition with UV280 detection.
